Kementec’s portfolio of substrates includes soluble Horseradish Peroxidase (HRP) substrates (TMB) and soluble Alkaline Phosphatase (AP) substrates (pNPP) and precipitating substrates enabling different substrate formulations for immunoassays.

Soluble HRP Substrates
Soluble Horseradish Peroxidase (HRP) Substrates
In a soluble chromogenic HRP assay setup, like an ELISA, the preferred substrate is TMB (3, 3', 5, 5'-tetramethylbenzidine). When developing immunoassays, choosing the right TMB is a vital step for optimizing the assay. Kementec offers a range of TMBs with different kinetics and sensitivities, enabling assay developers to choose the right TMB for their assay.
TMB Kinetics

All the TMBs are ECO-TEK, making them REACH, Annex XVII0 compliant and easy to transport with less restrictions. ECO-TEK includes:
- Free of BSA
- Free of NMP or other harmful organic solvents – 100% aqueous
- Free of toxic preservatives and hazardous stop solutions
The TMBs are characterized by outstanding sensitivity, stability and stop-stability, and very high lot-to-lot consistency. Offering a shelf life up to 4 years of the ready-to-use substrates enables Kementec products to meet all the requirements of assay manufacturers.
Lot-to-lot variation
In order to achieve high quality levels, rigorous QC standards ensure negligible lot-to-lot variations. 50 Lot-to-lot consistency, displayed in the example, shows the last 7 years productions of TMB PLUS2, which lie within a 20% interval.
Stop Solution Benefits
As we focus on environmentally safe products, Kementec substrates require low stop solution concentrations. The superior stop stability maintains the final OD level, as illustrated, where Kementec’s TMB X-tnd is compared to 1 competitor. The stop stability up to 60 minutes drops to no more than 5% in the high OD area.
Several of the TMBs are suitable for use in electrochemical assays that utilize sensor measurements.
TMB substrates are also available as unique color-coded products for traceable pipetting. The pink indicator makes manual pipetting traceable, while in automatic analyzers it offers a convenient tracking system for reagent supply. The color disappears after adding the recommended stop solution and does not interfere with the assay.
Corresponding diluents are available for TMB substrates in order to achieve a specific activity level.

Soluble AP Substrates
In a soluble chromogenic Alkaline Phosphatase (AP) assay setup, like an ELISA, the preferred substrate is pNPP (p-nitrophenylphosphate). Kementec offers two different pNPP solutions, which are mono-component and stable for up to 36 months.
pNPP liquid ready-to-use substrate is highly active. The substrate gives superior results due to its sensitivity, allowing very low detection limits.
pNPP ONE liquid ready-to-use substrate is incredibly stable. PNPP ONE shows superior background characteristics due to its high temperature robustness.

Precipitating Substrates
Kementec offers superior precipitating substrates for AP (BCIP/NBT) and HRP (TMB). The substrates are ready-to-use, robust and highly sensitive for applications like membrane blotting. The intense colored precipitates are incredibly stable and resist fading over time.
